Abstract
Conditions for the extraction of peroxidase from bean leaves were studied. Both the pH and the ionic strength of the extraction medium played an important role in the recovery of certain peroxidase isozymes. It was found that a major portion of the cationic peroxidase in the leaf tissue was sorbed to naturally occurring insoluble polymers at low ionic strength and neutral pH. These polymers seemed to act as ion-exchangers from which the enzyme could be progressively eluted by increasing sodium chloride concentration.
