Abstract
Evidence is presented, using the acidic dye acid fuchsin, that the Feulgen reaction is not based on acid-base mechanisms as suggested by a few authors. Various extraction, enzymatic and other procedures are used to show that the Feulgen reaction is specific for deoxyribonucleic acid when using either acid fuchsin or basic fuchsin in Schiff's reagent. There is no evidence here that any other nuclear component is stained in the Feulgen reaction.
