Optimization of the Formulation of Transdermal patches of Amiodarone

Abstract

The objective of the present study was to prepare transdermal patches of amiodarone using sustained release polymers enriched with permeation enhancer utilizing solvent evaporation method. The casting solution was prepared using hydroxypropylmethyl cellulose, ethylcelluose and Eudragit RS100 as the polymeric matrix. DMSO was used as the permeation enhancer and glycerol was used as the plasticizer. Optimization of the formulation was done using 23 factorial design followed by multiple regression analysis and surface response analysis of the release response to generate the polynomial equations to select the best combination of polymers. Hydroxypropylethyl cellulose and ethylcellulose were found to be most suitable copolymeric combination.
