Preparation and Characterization of Microwave Assisted Bionanocomposites for Enhancement of Physicochemical properties of Olmesartan

Abstract

Drugs which are fairly soluble in gastrointestinal (GI) medium show full oral absorption and, as a result, good bioavailability. In actuality, about 40% of medications are not soluble in water, which causes them to be absorbed slowly. This leads to uneven and insufficient bioavailability as well as GI toxicity. Therefore, improving a drug's solubility and, consequently, its oral bioavailability, is the most crucial stage in the drug development process, especially for oral dosage forms. Improving the solubility and dissolution rate of the weakly water soluble medication Olmesartan through the use of microwave-generated bionanocomposites was the goal of the current study. The bionanocomposites were prepared using natural polymer Moringa oleifera as compared with xanthan gum and guar gum. The prepared bionanocomposites showed enhanced solubility when compared xanthan gum and guar gum in trial batches. The prepared bionanocomposites were characterized by DSC, SEM, and XRD which indicates that crystallinity of olmesartan has been reduced significantly. Solubility study result helped for selection of ratio of drug and polymer. In-vitro drug release of prepared bionanocomposites shows better results when compared with marketed formulation. Accelerated stability study as per ICH guidelines of optimized batch was performed and it was found that formulation was stable for six months. Therefore, microwave assisted bionanocomposites using natural polymer can be a successful technique for enhancing the solubility of olmesartan.
