Abstract
This protocol describes a classical method for measuring cytoplasmic Ca2+ dynamics in Arabidopsis thaliana guard cells. The Förster (fluorescence) resonance energy transfer (FRET)-based genetically modified Ca2+ indicator Yellow Cameleon YC3.6, under the control of the guard cell-specific promoter GC1, is used for Ca2+ measurements.
