Abstract
For assay of serum aspartate aminotransferase activity the AutoAnalyzer II single-channel 340-nm colorimeter has been substituted in the AutoAnalyzer I system. The larger dialyzer requires proper balance between the liquid streams entering and leaving the flow cell, and between air-bubble size and total flow rate. For precise air metering, an air-bar has been improvised. Analysis rate is 60 per hour, the samplewash ratio is 1:2. Through-put time is 15 min. The reaction is linear to 200 Karmen units without serum blanks. Sera were analyzed by our hybrid system, by an automated Transac procedure, by Henry’s manual 340 nm kinetic method, and by the 340-nm SMA-12 method. The results were not significantly different. Within-run precision at three concentrations is 2.13.8% (coefficient of variation). Day-to-day precision CV is 4.3%. Interaction is 2 to 4%.
