Abstract
In this study, we present centrifugal flow disk to isolate single cell and culture it from single cell simply and rapidly by combining the aligned microchambers and centrifugal force. In order to determine activity of single-cell, a centrifugal flow disk was designed and made of polydimethyl-siloxane (PDMS), which has gas permeability. On the disk 24 microchannels were arranged and along each channel 530 microchambers were aligned. To confirm the activity of isolated cell, cell suspensions of Eschericia coli, baker's yeast, Jurkat cell, and NIH3T3 cell are isolated by the rotation of the disk at 3000 rpm, 30 sec. After the isolation, cells of E. coli and baker's yeast were cultivated from the isolated single-cell to large amount of cells after 24 hours of incubation. As for Jurkat cells and NIH3T3 cells, two or three times of cell division were performed easily in these disks without medium exchange.
