Chimeric a-subunit isoforms generate functional yeast V-ATPases with altered regulatory properties in vitro and in vivo

Abstract

V-ATPases are highly regulated proton pumps that are localized and regulated by a-subunit isoforms. Chimeric a-subunits containing parts of different isoforms were designed, their interactions with regulators analyzed, and their function under different stresses determined. One chimeric V-ATPase is as active as the wild-type enzyme but shows distinct regulatory properties in responses to stress.
