ABSTRACT
The conversion by a corpus luteum of pregnancy of pregnenolone-7-3H and progesterone-4-14C to oestradiol in 1% yield has been reported. The oestradiol was characterized by solvent partition, chromatography, enzymatic conversion to oestrone, methyl ether formation and crystallization after carrier addition. A similar ratio of 3H to 14C was found in the starting materials, in the 17-hydroxyprogesterone intermediate and in the end product oestradiol. No evidence was found for a significant alternate pathway of oestrogen formation via Δ5-3β ol intermediates in this tissue.
