Abstract

ABSTRACT A method for the simultaneous determination of free cortisol and of dehydroepiandrosterone and androsterone conjugates in human plasma is described. Chloroform extraction, subsequent paper chromatography and alkaline tetrazolium-blue reaction were used for measuring free cortisol values. For splitting of 17-oxosteroid conjugates, simultaneous acid hydrolysis and benzene extraction are employed. An extensive purification of the extract is achieved by thin-layer chromatography. Paper chromatography is used for separation, and in situ Zimmermann-reaction applied for quantitation of individual 17-oxosteroids. The control of the method, the normal values and the plasma steroid level in different endocrine disorders are presented.
