Subtotal synovectomies were performed on the knee joints of rabbits, and 15 hours before being killed at daily intervals, 0 2 mc. 35S-labelled sodium sulphate was administered to each animal intravenously. Up to 15 days, the maximum period studied, the operated knees showed no decrease in the uptake by the articular cartilage of the labelled sulphate radical, as measured by autoradiography. Compared with the control knees, there was a suggestion that the superficial layers of cartilage in the synovectomized knees showed increased uptake of sulphate. This was considered to indicate that, during the period studied, synovectomy produced no impairment of the ability of the chondrocytes to absorb and utilize sulphate. In a further series of animals in which synovectomized knee joints were stained with toluidine blue after being killed at intervals up to 100 days, absence of metachromasia, in varying degree, was noted during the period of synovial regeneration. This is considered to be due to diffusion of a cathepsin or plasmin from the joint cavity into the cartilage, with subsequent enzymatic degradation of the protein-polysaccharide ground-substance, while the increased uptake of sulphate by the superficial layers is thought to represent an attempt by the chondrocytes to replace this ground substance.
