Abstract

We introduced a nomenclature for split inteins and engineered mini-inteins. 21 new split inteins were created based on the four intein structures.trans-Splicing and cross-activity of 24 split inteins were systematically compared. >75% of the split inteins were active and >90% of the combinations were orthogonal.
