Abstract
This study aimed to investigate the effect of Troxerutin on TGF-β-induced fibrosis of renal tubular cells and whether it can participate in the activation of MG-53. After induction of HK-2 cells with TGF-β 100 ng/ml for 24, 48 and 72 hours, the level of inter cellularization
was detected by western blot. After treatment with different concentrations of Troxerutin, The expression of E-cad, N-cad, -SMA, collgen-I and FN were detected by western blot, and the expression of N-cad and -SMA were detected by immunofluorescence. Furthermore, the interference plasmid of
MG-53 was transfected into HK-2 cells. The expression of MG-53, E-cad, N-cad, α-SMA, collgen-I and FN expression were detected by western blot. TGF-β-treated HK-2 cells increased inter cellularization and fibrosis, and Troxerutin attenuated TGF-β-induced cell cytoplasmic and
fibrotic protein production. TGFinhibits the expression of MG-53, while Troxerutin increases the expression of MG-53, and reduces TGF-β-induced interstitial and renal fibrosis by increasing MG-53. These results suggested that troxerutin attenuates TGF- β-induced renal cellular fibrosis
in HK-2 cells by activating MG-53. This study provided novel insights into the mechanisms of troxerutin in the protection of the renal.
