Abstract
The effect of urea upon the activity of cod muscle cathepsin on hemoglobin substrate was studied.The apparent activation of the enzyme, found when urea was present, either added to the hemoglobin substrate before incubation or added at the start of the enzyme incubation with substrate, may be explained as an increase in the solubility of the enzyme reaction products in the presence of urea when trichloroacetic acid is added to end the incubation. This was shown by addition of urea together with trichloroacetic acid at the end of incubation. This resulted in activity measurements higher than any when urea was present during the incubation, and demonstrated that the effect of urea is to inactivate rather than activate the enzyme.
