Abstract
A new method for preparing eggs of the migratory grasshopper, Melanoplus sanguinipes Fab., for serial sectioning is described. The fixative is Carnoy's fluid; dehydration and clearing are accomplished in dioxane and benzene. After infiltration and embedding, tissues are exposed on the microtome and then soaked in a detergent solution for 4 hours or longer. The entire procedure from fixation to sectioning can be completed in 16 to 20 hours, but one or more overnight stops may be included.
