Tree peony is a shrub plant of great economic value due to its large and beautiful flowers, seeds containing much oil, root used as medicine. Understanding the genetic diversity and relationship of the tree peony cultivars is of great significance in facilitating breed and cultivar maintenance. In this study, we analysed genetic diversity and clustering of eight cultivated tree peony (Paeonia ostii) lines in Democratic People’s Republic of Korea using RAPD and SRAP primers. Dendrogram analysis indicates that eight lines were divided into two groups (Cluster I: D1, D2, D3, Cluster II: N1, N2, N3, N4, N5), which is consistent with clustering based on seed productivity. Clustering analysis based on STRUCTURE analysis and PCA analysis supports the phylogenetic analysis results. The RAPD and SRAP primers used in this study could be used to facilitate breeding, cultivar identification and maintenance to increase seed productivity.
