Somatic embryogenesis as a way of Siberian pine biodiversity preservation: initiation

Abstract

Abstract The article reports about the results of an experiment to initiate somatic embryogenesis in Siberian pine in 2019. The object of the study was trees from the forest seed orchards of Berdsk forestry in the Novosibirsk region. Immature cones were collected at the stage of early embryogenesis. 5 culture media compositions and 6 genotypes were tested. Intact and dissected megagametophytes were used as explants. Two stably proliferating embryogenic lines were obtained. It has been proven that the genotype and composition of the nutrient medium influence on the initiation of somatic embryogenesis. The most competent genotypes and effective nutrient media for somatic embryogenesis of Siberian pine were revealed. It has been established that dissected megagametophytes are promising to use as explants for obtaining somatic embryogenesis in Siberian pine.
