Antioxidant Activity of Sappan Wood Extract (Caesalpinia sappan L.) As An Adjuvant and Substitution in Blood Serum of Rats (Rattus norvegicus) Iron Overload Model

Abstract

Abstract Sappan (Caesalpinia sappan, L.) contains various antioxidant compounds such as brazilin and flavonoids that able to reduce superoxide anion radicals, reduce hydroxyl free radicals and chelate iron ions. This study aims to obtain the antioxidant activity of sappan wood extract which is given adjuvantly and substitution in blood serum of overload iron model rats. The research method used is experimental laboratory with a completely randomized design. The test animals were used 70 male rats aged 7 weeks weighing 200 grams and divided into normal, negative control, comparison, and experimental group. The adjuvant group used sappan wood extract at doses of 50, 100, 150, and 200 mg/kgBW. The subtitution group administred various percentages (25%, 50%, 75%, and 100%) of the sappan wood extract dose of 200 mg/kgBW. Data obtainedwere analyzed using one way ANOVA and Duncan’s test at 95% confidence level. The results showed that the administration of sappanwood extract at a dose of 50 mg/kg BW as an adjuvant and a dose of 200 mg/kg BW (100%) by substitution could provide better antioxidant activity against iron-excessed rat blood serum. This can be seen from the decreased activity of SOD, and levels of MDA and increased activity of GPX.
