Am J Pathol 2001, 158:1677–1683. This study showed an accumulation of genetic alterations with increasing PanIN grade. Moreover, the data indicate that allelic loss is the first hit in the biallelic inactivation of the p53 and DPC4 tumor suppressor genes. Taken together, the data support a tumor progression model for PDAC.

This paper presents the PanIN classification, which standardizes terms to cover the various changes in the pancreatic duct system that seem to be relevant to the development of PDAC.
