Pterostilbene regulates endothelial cells activation and expression of adhesion molecules through HIF-1α in cerebral vessels with hypoxic injury

Abstract

This study assessed mechanism of pterostilbene in regulating endothelial cells activation and expression of adhesion molecules in cerebral vessels with hypoxic injury. These endothelial cells in cerebral vessels were cultivated in vitro and model of hypoxic injury was prepared. Cells were assigned into control group, model group and group with pterostilbene. The action of pterostilbene on proliferative and apoptotic activity was tested. Expression of HIF-1α, VCAM-1 and ICAM-1 was detected, and change of SOD activity and MDA content and VEGF level were tested. Results revealed that, activity of endothelial cells and proliferation was increased in the pterostilbene group, while apoptosis and expression of HIF-1α were reduced. The expressions of VEGF and SOD activity were increased, while that of VCAM-1 and ICAM-1 were reduced, including the MDA content. The expression of HIF-1α was reduced while that of VEGF was increased by pterostilbene. Redox activity was also improved and adhesion molecules was restrained. Endothelial cells activation in cerebral vessels with hypoxic injury was thus improved by pterostilbene.
