Abstract
Objective To study the expression of Bax, Bcl-2 and Caspase-3, three apoptosis-related proteins after skeletal muscle injury in rats, and explore their application value in distinguishing antemortem and postmortem injuries.
Methods 42 healthy adult Sprague-Dawley rats were randomly divided into control group, antemortem injury group and postmortem injury group. The gastrocnemius muscle injury of the right hind limb of rats was made. The feasibility of the model was verified by HE staining. The mRNA expression was detected by Q-PCR. The expression of Bax, Bcl-2, and Caspase-3 proteins was detected through Western Blot, the data were centralized and standardized respectively, and orthogonal partial least square-discriminate analysis (OPLS -DA) of antemortem and postmortem injuries was established.
Results HE staining cannot accurately distinguish between antemortem and postmortem injuries; The changing trend of Caspase-3 mRNA expression is consistent with the protein expression trend; the protein expression levels of Bax, Bcl-2, and Caspase-3 were centered and standardized and analyzed through OPLS -DA. The model can distinguish antemortem injury or postmortem injury samples.
Conclusion The expression of Bax, Bcl-2 and Caspase-3 proteins in local tissues of rat skeletal muscle injury can be used to differentiate between antemortem and postmortem injuries.
